BMS Shares Positive Phase 3 Results for Oral Mezigdomide in Multiple Myeloma

Bristol Myers Squibb has announced positive interim results for its SUCCESSOR-2 phase 3 trial of oral mezigdomide to treat relapsed or refractory multiple myeloma (RRMM).

The treatment is part of the BMS targeted protein degradation (TPD) platform, which uses immunomodulatory drugs (IMiDs) to therapeutically degrade proteins that were previously considered impossible targets. The program relies on a three-pronged approach, with CELMoD agents, ligand-directed degraders (LDDs), and degrader antibody conjugates (DACs). Oral mezigdomide is a CELMoD — the second to achieve positive phase 3 study results for the company.

In the clinical trial, participants received oral mezigdomide alongside carfilzomib and dexamethasone (MeziKd). The treatment led to a statistically-significant improvement in progression-free survival (PFS) compared to just carfilzomid and dexamethasone (Kd). MeziKd also a safety profile comparable to Kd.

The trial data underscore the potential of MeziKd as an oral regimen that could address a key unmet need for patients who relapse after being previously exposed to anti-CD38 and lenalidomide.
